Reduction of IL-5 receptor on BAL eosinophils 18 h and on blood eosinophils 42 h after allergen challenge

Abstract

In asthma, IL-5 plays an important role in the chemotaxis of eosinophils. Its receptor is expressed on eosinophils, basophils, but not on neutrophils or monocytes. In this study we investigated IL-5 concentrations in BAL and IL-5-receptor expression on peripheral blood and BAL eosinophils 18h and 42h after segmental allergen provocation in vivo.
An individually standardized dose of allergen or saline was instilled into different segments during bronchoscopy in 16 subjects with mild, allergic bronchial asthma. Segments were lavaged 10 minutes, 18 or 42 hours after allergen challenge or saline instillation. IL-5R was measured by flow cytometry and IL-5 concentrations in BAL-fluid by ELISA.
IL-5R expression was significantly reduced on BAL-eosinophils 18h after allergen challenge, but not after 42h while on blood eosinophils the reverse was found. IL-5-concentrations in BAL were significantly elevated 18h and 42h after allergen challenge.
We conclude, that IL-5 may have a down regulating effect on its own receptor in vivo.
