Interleukin-4 and interferon-gamma production by allergen and PPD stimulated T-cells in the presence of IL-12 and IL-18

Abstract Interleukin-18 augments IL-12 induced IFN-gamma synthesis from T cells. However, recently it has been shown that this cytokine may also increase IL-4 synthesis from NKT cells and enhance Th2 responses in a murine model of allergic asthma. The aim of the study was to investigate whether IL-18 affects cytokine production by allergen or nonallergen stimulated T cells and whether it synergizes with IL-12 in CD30 receptor expression. Methods: Peripheral blood mononuclear cells were isolated from healthy or atopic patients. Cells of healthy subjects were stimulated with PPD and those from atopic patients with the allergen that caused positive skin prick reaction. The optimal concentrations of antigens used to stimulate cells were derived from proliferation test results. Cells were stimulated for 10 days and refed with antigen presenting cells, specific antigen and cytokines or anti-CD3 antibodies and cytokines. The production of IL-4 and IFN-gamma was determined by standard ELISA procedure in supernatants collected on the 3-rd day after stimulation to coincide with the optimal CD30 expression. Results: Restimulation of allergen specific T cells and PPD cells in the presence of IL-18 significantly increased IL-4 production but had no effect on CD30 expression. IL-18 had no effect on IFN-gamma synthesis by PPD and IL-12 stimulated cells what we explain by the maximal production of this cytokine by cells stimulated by PPD alone. This cytokine however lowered IL-12 induced IFN-gamma production by allergen stimulated T cells. Conclusion: Our results add another evidence for Th2 like activity of IL-18 and show that this cytokine may affect not only allergen but also nonallergen specific T cells.
