The role of interleukin-10 and interleukin-8 in the evolution of asbestosis

Abstract

To investigate about sensitive indicators of exposure or early changes of lung damage in evolution of asbestosis, 25 subjects exposed to asbestos underwent bronchoalveolar lavage (BAL).
The subjects (24 males and 1 female, mean age 57.4 years) were divided according to the extent and duration of their exposure, to the concentration of asbestos fibres in the BAL and to Computerized tomography (CT) of the chest findings, using the I.L.O.classification (0/0 and 0/1pl vs 1/0 and above).
The BAL was examined for cytological parameters, lymphocyte subpopulations, cytokines in the supernatant (IL8, IL10, IL12 and MCP-1) and the present and quantity of asbestos fibres. No correlations were showed comparing exposure index with BAL parameters.
In the group of subjects with a 0/0-0/1-pl radiological profile, a higher percentage of lymphocytes(p=0.019),T total (CD3) (p=0.02)and T helper (CD4)(p=0.05)lymphocyte subsets was noted in the BAL. On the other hand, an increase of neutrophils was shown in subjects with marked pulmonary fibrosis. The percentage of neutrophils was inversely correlated with VC(r=-0.41, p<0.02) and pO2(r=-0.3, p<0.05).
Moreover we found a negative trend of IL-10 BALF concentration as the radiological picture worsened, and a positive correlation between lymphocytes and MCP-1 (r=0.55,p=0.009) and between neutrophils and IL8 (r=0.43,p=0.038) in bronchoalveolar lavage.
This research suggests a pathogenetic role of IL10 and IL8 in the evolution of asbestosis, but more observations are needed.
