Pi phenotypes of antiprotease α1 -antitrypsin and log dose slope of histamine responsiveness

Abstract Objective. To study the association between histamine bronchial responsiveness (Yan method) and Pi phenotypes of antiprotease α1 -antitrypsin (α1 -AT) in 1807 male rurals, aged 18.6±]2.0 years, mean±]SD.Method. Histamine chloride was given up to a dose of 1.43 mg. and log dose slope (LDS) was calculated as: log10((FEV1start - FEV1end ) * 100/ (FEV1start * dose) + 1). In subjects with α1 -AT serum concentration [lte] 22 μM phenotypes was tested and subjects with higher concentration were classified as MMconc . Phenotyping was performed by means of isoelectric focusing. The different Pi-groups were collapsed into three: MM: MMconc , MM and MX; MZS: MS and MZ, and SZ: SZ, ZZ and SS. The X represent any other allelic type.Results. The LDS (mean±]SD) was 0.72±]0.49 for the 1621 subjects with MM Pi-group. The LDS was 0.78±]0.44 for the 177 subjects with Pi-group MZS and the LDS was 1.04±]0.82 for the 9 subjects with Pi-group SZ (linear regression: Β 0.081 (0.032), p=0.01).Conclusion. LDS(FEV1 ) of histamine responsiveness was associated to Pi-phenotypes of α1 -AT. Subjects with rare alleles seems to be the most responsive.
