Abstract

There are controversies regarding airway responsiveness to β-agonist in asthma. Epithelial shedding of the airways is a pathological feature of asthma. Therefore, in the present study ,the effect of epithelial denudation of guinea pig tracheal chians in responsiveness to isoprenaline and also .β-adrenergic receptor blocked was studied.
The responses of tracheal chains of control, epithelial denuded (for each group n=10) and epithelium denuded+epithelial cell (n=6) to cumulative concentrations of isoprenaline (I) in the absence and presence of 10 nM propranolol were measured, and the effective concentration of I causing 50% of maximum response (EC50 I) was obtained. The propranolol blockade (CR-1) was calculated by: (post propranolol EC50 I/EC50 I)-1.
EC50 I in epithelial denuded group was significantly lower than that of control group (0.22±]0.07 vs 0.55±]0.04 μM, p<0.05). The propranolol blockade (CR-1) was also significantly higher in epithelial denuded trachea than that of control group (6.57±]0.65 vs 2.34±]0.42, p<0.001). EC50 I of the epithelium denuded+epithelial cell group was also significantly lower than that of control group (p<0.05), but was not significantly different with that of the epithelium denuded group. However, (CR-1) in this group was not significantly different with that of control, but was significantly lower than that of the epithelium denuded group (p<0.01).
The results of these studied indicated an increased tracheal response to β-adrenergic stimulating drug and enhancement β-adrenergic blocked by propranolol.
