Abstract Objective: To investigate the morphologic characteristics and its effect on airway responsiveness, and to observe the intervention effects of glucocorticoids and cromolyn sodium to airway remodeling. Method: (1) sensitized guinea pigs were repeatedly exposed for periods of 4, 6 or 8 wk to aerosolized ovalbumin(OA) or of 4wk to aerosolized saline. (2) The morphologic parameters were measured in large, medium and small airway sections including the areas of inner wall, outer wall, smooth muscle and fiber tissue. (3) Bronchial provocation tests were conducted and the resistance of airway was recorded.(4)Glucocorticoid and cromolyn sodium were given to the animals for 4wk and the whole exposure to OA was 8wk, and the effects of both drugs were evaluated. Results: (1) The areas of inner wall and fiber tissue in large airway and the area of smooth muscle layer of small airway were significantly increased after 8 wk of OVA exposure. (2)A 4wk exposure to OVA induced an increase in airway responsiveness, but the increase disappeared upon prolonged exposure. (3) The area of airway fibrosis was decreased, but there was no effect on the thickness of smooth muscle after treated by glucocorticoid. Allergen-induced airway fibrosis was partially reversed after cromolyn sodium was used. Conclusion: (1)Airway remodeling included the thickening of the airway, the proliferation of the fiber tissue, the hyperplasia of the smooth muscle (2) More prolonged exposure to OVA resulted in an increase followed by decrease in airway responsiveness. It indicated that airway remodeling had a protective role in bronchial responsiveness.(3) Both glucocorticoid and cromolyn sodium could partially reverse airway fibrosis, but neither glucocorticoid nor cromolyn sodium has effect on smooth muscle.
