Comparison of amosite and wollastonite fibre effects on selected BAL parameters and lung tissue injury-time and dose dependence

Abstract The problem of the effects of industrial fibrous dusts on the respiratory tract remains topical.Aiming to determine the influence of dose and exposure time on lung injury, we investigated inflammatory and cytotoxic bronchoalveolar lavage (BAL) parameters (number of leukocytes (Le), alveolar macrophages (AM); cell differencial; total amount of protein (TAP); phagocytic activity and viability of AM; activity of lactate dehydrogenase (LDH), acid and alkaline phosphatase (AcP, AP) and cathepsin D) and histology of lungs after intratracheal instillation of amosite - asbestos and wollastonite - asbestos substitute fibres (2 and 8 mg) after 4 and 16 week exposure in rats. We found:1) Wollastonite - did not significantly influence BAL parameters and lung structures in comparison with the control. We suppose that absence of wollastonite effect on mentioned parameters might be consequence of its short half time of elimination (low biopersistence); 2) Amosite - most of evaluated BAL parameters were significantly changed after both dose exposure not only in comparison with controls but also with wollastonite groups (significantly increased: number of Le, TAP, LDH - p < 0.01; significantly decreased: % of total AM - p < 0.05; phagocytic activity and viability of AM - p < 0.01;). The adverse effect of amosite was more expresive after 8 mg instillation - mainly in cytotoxic parameters (moreover increased AcP, AP - p < 0.01, cathepsin D - p < 0.001). These fibres caused serious interstitial fibrosis and progresive inflammation with evident time and dose dependence. The work was supported by an E.U. grant, contract, No. QLK4-CT-1999-01629.
