Long-term effects of nebulized LPS in a mouse model of acute lung injury (ALI)

Abstract
Background
: Several animal models were developed to elucidate the mechanisms of acute lung injury (ALI). One of the most frequent used models is the LPS-induced ALI by intratracheal instillation. Some short-term models (up to 48 hours) have used nebulized LPS. Few experimental models are suitable to investigate the long-term evolution of pulmonary changes in ALI.
Aim
: To evaluate the long term (five weeks) effects of nebulized LPS in a mouse ALI model.
Methods
: ALI was induced by nebulized LPS (3mg/ml). BALB/C mice with or without ALI were euthanized 5 weeks after the ALI induction. Lungs, blood and BALF were collected. White blood cell count and total BALF cell count were obtained. Lungs were evaluated using stereological methods. We estimated total and volume densities, surfaces and surface densities of lung compartments (septum, alveolar space, airways, and blood vessels) and the septum arithmetic mean thickness.
Results
: The blood concentration of leucocytes (p=0.0001), neutrophils (p=0.0001) and lymphocytes (p=0.0001) were significantly increased in the ALI group. The cell count on BALF also showed an increase in the ALI group (p=0.002). The total volume of the lungs (p=0.011), septum (p=0.0001), alveolar space (p=0.029), blood vessels (p=0.0001) and the septum arithmetic mean thickness (p=0.023) were greater in the ALI group. Decreased airway lumen volume (p=0.021) was also observed in the ALI group.
Conclusion
: The nebulized LPS-induced ALI model is feasible and shows ALI structural and inflammatory features even after five weeks of exposure.
