Calcium channel activity in lowlanders and in highlanders with pulmonary hypertension due to chronic obstructive pulmonary disease

Abstract Increase in intracellular Ca2+ content mediates hypoxic pulmonary vasoconstriction resulting in increased pulmonary artery pressure. We investigated calcium channel activity on the blood thrombocytes in highlanders and lowlanders with COPD and pulmonary hypertension (PH). Intracellular Ca2+ content was measured with a probe Quin-2. Pulmonary hemodynamics was evaluated during righ heart catheterization at baseline while room air breathing and during hypoxic challenge (breathing FiO2=0.1 for 30 min). Results are presented in the table. Basal Ca2+ content demonstrated was significantly increased in highlanders with COPD and PH. There was no difference in basal calcium content between healthy lowlanders and lowlanders with COPD and PH. However induction with ADP markedly increased intracellular Ca2+ content on the blood thrombocytes in patients with PH more significant in highlanders. Thus increased calcium channel activity in patients with hypoxic PH is one of the main mechanisms underlying hyperreactivity of pulmonary vascular bed, which contributes to the maintenance and progression of PH. Legend.
