Abstract
Pulmonary Emphysema is the lung tissue destructive variant of a group of chronic lung diseases (COPD). Although smoking is by far the most common cause of Emphysema, only approximately 15% of chronic smokers develop Emphysema, which strongly indicates a genetic susceptibility for the development of this disease. Using an oligonucleotide microarray technology (Affymetrix GeneChipâ) we characterized the tissue gene expression profile in 6 patients with severe Emphysema and compared the gene expression profile in lung tissue samples from patients with α1-AT deficiency (n=4) and 6 patients with histologically normal lungs. To confirm the microarray data we performed quantitative RT-PCR. The gene expression profile in lungs from patients with α1-AT deficiency was generally similar to the expression profile in emphysematous lung samples obtained from patients without α1-AT deficiency. Differences in the gene expression profile were noted when lung tissue samples from patients with severe emphysema were compared to histologically normal lung samples. Genes which affect lung structure maintenance, growth/proliferation, apoptosis, inflammation, protease/antiprotease imbalance, energy metabolism, protein processing were differentially expressed in emphysematous compared to normal lungs. We conclude that the microarray gene expression technique is a new and useful molecular tool that provides novel information pertinent to a better characterization and understanding of the pathobiology of pulmonary Emphysema.
