Relation of cytokine expression profile of T-cells from blood and bronchoalveolar lavage fluid with HRCT pattern in fibrosing interstitial lung diseases

Abstract

Background: T-cells are potent source of cytokines, that may play role in pathogenesis of interstitial lung diseases (ILD). High resolution computed tomography (HRCT) is an nonivasive method suggested to reflect both extent of fibrotic and inflammatory changes. Correlation between radiologic pattern of the disease and expression of proinflammatory/profibrotic cytokines might suppose our hypothesis, that used HRCT scoring system (1) is a powerfull tool of disease activity assessment.Methods: Twenty prospectively enrolled patients underwent bronchoscopy with BALF, HRCT and collection of blood samples. Spontaneous and stimulated expression of IL-2, IL-4, IL-12 and INFg by CD4+ and CD8+ T-cells from BALF and PB were measured by flow cytometry before and after nonspecific stimulation (PMA, Ionomicin). Relations between variables were estimated by Spearman correlation coeficient.Results: Positive correlation between HRCT alveolar score and spontaneous expression of IL-2 in PB by CD8+cells (r=0,54, p=0,01) was observed. No correlations of either spontaneous or stimulated expression of cytokines by BALF and PB T-cells with HRCT score were found.Conclusion: We did not prove relevant correlation between HCRT score and expression of IL-4, IL-12, INFg by T-cells obtained from spontaneous/activated PB and BALF.
