Long term follow-up of Greek patients with pulmonary sarcoidosis. Relation to transforming growth factor-beta genetic profile

Abstract

Objective: The aim of the present study was to investigate the development of pulmonary sarcoidosis and its possible correlation with transforming growth factor-beta (TGF-b) gene polymorphisms.Methods: Sixty-five patients with biopsy proven pulmonary sarcoidosis , 24 males and 41 females, mean age 46.3±10.8 years, 15 with acute/self remitting and 50 with chronic disease and 19 healthy controls, mean age 37.8±6.4 years, were included in the study. All patients were recruited from the Out-Patient Clinic for Sarcoidosis, Pulmonary Department, Aristotle University of Thessaloniki and had a mean follow-up time of 9.1±6.4 years. Classification by radiological stage was made both currently and at initial diagnosis. For the immunogenetic study DNA was extracted from the patients and controls peripheral blood samples. Single nucleotide polymorphisms (SNPs) in TGF-b2 (rs 1891467) and TGF-b3 (rs 3917200) were investigated by real time PCR.Results: A change in stage classification was observed over time. 35 patients showed a complete remission, 15 of them spontaneously. The distributions of allele carrier and genotype frequencies of TGF-b2 and TGF-b3 in patients and controls did not deviate from the Hardy–Weinberg equilibrium.Genotype frequencies analysis did not show any significant differences between patients and controls.A statistically significant difference was observed between decreased frequency of C-Allele in the TGF-b3 and acute course among male patients (p:0.018). There was no association between the TGF-b2 or -b3 genotype and radiographic stage.Conclusions: TGF-b3 gene polymorphisms might play a role in the development of pulmonary sarcoidosis.
