The activity of metalloproteinase-2 and metalloproteinase-9in cerebral arteries in experimental smoking

Abstract Aim: to examine the activity of metalloproteinase-2 (MMP-2), metalloproteinase-9 (MMP-9)in cerebral arteries in the experimental smoking.The model is based on smoking protocol H. Zheng on Wistar rats (group 1). The animals had 2 hours exposure to smoke tobacco for 6 months. Control group of rats was kept in standard conditions (group 2). The histologic samples of rat's brain were produced after euthanasia. The localization and activity of MMP- 2 and MMP- 9 of in cross sections of vessels in the dura mater were analyzed by immunohistochemistry method with immunoperoxidase mono-and polyclonal antibodies against MMP- 2 and MMP- 9. Activity of the reaction in cross sections was measured in units of optical density. Density immunohistochemistry reaction in 1 cm3 of brain tissue was evaluated by Jenaval Carlzeiss Jena microdensitometer. The results were processed by Adobe Photoshop 7.0 and Image J computer programs. Statistical confidence was estimated with Mann- Whitney index. Results: Significant descension of activity and the specific density of the precipitate distribution of MMP-2 in wall of cerebral arteries of rats group 1 was estimated, respectively, to 12,46 +6,1 and 164,3+70,7 compared with group 2 (25,17+ 9,9 and 344,2+88, p<0,05). Also increasing of specific activity and the distribution density of MMP-9 was determined, respectively 11,6+1,25 and 132,6+6,8 in group 1, compared with group 2 (6,97+0,9 and 41,9+ 1,8; p<0,05).Conclusion: Descension activity of MMP-2 and increasing activity MMP-9 in wall of cerebral arteries was estimated in experimental tobacco smoking, which may be reflects with dismetabolism of connective tissue.
