Differential bronchoalveolar expression of candidate microRNAs in chronic obstructive pulmonary disease, asthma bronchiale and pulmonary sarcoidosis

Abstract Background: Lung inflammation is a characteristic hallmark of COPD, asthma bronchiale (AB) and pulmonary sarcoidosis (S). Profiles of inflammatory molecules are regulated posttranscriptionally by microRNA (miRNA). There is little information on bronchoalveolar (BAL) miRNA expression in vivo and on possible differences between aforementioned diseases which differ in their pathobiology. Method: Relative expression of 5 candidate miRNAs (mir-452, mir-129-3p, mir-146a, mir-21, mir-31) in BAL cells from non-smokers with COPD (n=10), AB (n=13), S (n=42) and healthy control subjects (n=8) was determined by Real-Time PCR. Results: Comparing with controls, BAL expression of mir-452 (p COPD =0.01, p AB =0.01, p S <0.001) and mir-129 (p COPD =0.03, p AB =0.01, p S <0.001) were decreased in all 3 investigated diseases. Mir-146a (p AB =0.04, p S =0.002) and mir-21 (p AB =0.01, p S =0.001) were decreased in AB and S, while mir-31 alone was decreased only in sarcoid BAL (p =0.001). Other two miRNAs (mir-452, mir-129) were downregulated in S compared to AB and COPD (p =0.001). Conclusion: The differences in BAL miRNA expression between the investigated pulmonary diseases may contribute to variation of inflammatory response in their pathomechanisms.
