Surface phenotyping of peripheral blood stem cell-derived human basophils: comparison with blood basophils

Abstract

We have established a culture method for in vitro expansion of basophils from peripheral blood stem cell (PBSC) harvests. We cultured mononuclear cells from granulocyte-colony stimulating factor (G-CSF)-mobilized PBSC harvests, which were collected from patients with malignant lymphoma and lung cancer. After 3 weeks of culture in the presence of interleukin (IL)-3, 35-80% of the cells were identified as basophils. These cells (PBSC-derived basophils) were similar to peripheral blood basophils morphologically when examined by light and electron microscopies. They expressed both FcεRI and FcγRII abundantly. Moreover, we could obtain highly purified basophils (purity > 95%) by the effective removal of contaminating lymphocytes using immunomagnetic beads.
Flow cytometric analysis showed that PBSC-derived basophils expressed CD11a (LFA-1α), CD11b (CR3), CD11c (CR4), CD18 (LFA-1β), CD25 (IL-2Rα), CD29 (VLAβ1), CD49d (VLAα4), CD49e (VLAα5), CD54 (ICAM-1), CD116 (GM-CSFRα), and CDw123 (IL-3Rα). On the other hand, CD3, CD7, CD14, CD15, CD16 (FcγRIII), CD23 (FcεRII), CD36, CD45RA, CD64 (FcγRI), and HLA-DR were negative.
These findings support that PBSC-derived basophils express a close surface phenotype to that of peripheral blood basophils in terms of surface membrane receptors for immunoglobulins and cytokines as well as adhesion molecules. Thus PBSC-derived basophils will be a useful tool for the analysis of basophil functions in allergic inflammation such as bronchial asthma.
