BCG effect in allergic response to ovalbumin in genetically selected strain of mice (IV-A)

Abstract

Asthma is a multi-factorial and complex disease. The aim of this study is to check if BCG treatment in genetically selected mice down-regulates lung allergic response to OVA.
Genetically selected mice for high (HIV-A) or low (LIV-A) response to antibody against heterolog erythrocytes (IV-A selection) were treated with BCG i.m. or s.c., and immunized with OVA associated to BCG. 48h after antigen challenge, we measured anaphylactic antibodies IgE and IgG1 anti-OVA by PCA technique, and eosinophils in tissue and BAL and pulmonary conductance and compliance.
IgE and eosinophils in BAL were higher in HIV-A mice (1:400) than LIV-A (1:106); IgG1, eosinophils in tissue and other cells in BAL were similar in both strains; conductance and compliance were lower in immunized groups compared to saline. The animals that were injected with BCG showed lower IgE but not IgG1 antibody titles, and also reduced eosinophils infiltration in tissue and BAL. BCG down-regulates lung allergic response to OVA. The genetic selection per se was not enough to alterate this response in this experimental model of hypersensitivity in vivo.
