Abstract

Interleukin-5 (IL-5) plays a key role in the development of airway inflammation in asthma. IL-5 is produced mainly by T cells. To observe the association of the activation of T cells and expression of IL-5 mRNA, co-expression of CD25 and IL-5 mRNA were assessed with methods of immunocytochemistry and in situ hybridization double staining.
We found that there was a strong tendency to co-expression of IL-5 mRNA together with CD25 in T cells. 57.8+/-11.3% and 25.3+/-10.7% T cells from allergic mice were CD25(+)/IL-5 mRNA(+) and CD25(-)/IL-5 mRNA(-) respectively. CD25(-)/IL-5 mRNA(+) and CD25(+)/IL-5 mRNA(-) T cells were only 10.2+/-4.7% and 6.7+/-3.6% respectively. CD25(+)/IL-5 mRNA(+) T cells from allergic mice treated by dexamethasone decreased markedly (11.0+/-2.2% vs. 57.8+/-11.3%, p<0.01), and CD25(-)/IL-5 mRNA(-) T cells increased significantly (76.4+/-7.5% vs. 25.3+/-10.7% , p<0.01). These results indicate that expression of IL-5mRNA associate strongly with activation of T cells.
