Abstract

RATIONALE One of the main problems of TB control is its global dissemination. The ability of M. tuberculosis to spread between countries, therefore has important public health implication. Typing of M. tuberculosis strains by molecular methods can help to determine the ways of dissemination of M. tuberculosis from higher to lower prevalence areas.
METHODS We had used IS 6110- PCR typing method that allow us to amplify the flanking sequences on both sides of IS6110 by a single primer GACTCACCGGGGCGGTTCA.
RESULTS According to data of IS6110-PCR typing, M. tuberculosis strains divided into 4 groups of clusters and 70% of isolates get into one of the cluster groups. Groups of clusters N1 and N2 are the most frequent. We have determined correlation between each group of clusters and TB diagnosis ( infiltrative form of TB , disseminative form of TB , fiber-cavern form of TB). There are some peculiarity in clusterisation of TB isolates from fiber-cavern form of TB. The most frequent was type N1 (66%) and there were no isolates with type 2 and 3. On the other hand isolates from disseminative form of TB were type N2 (33%) and N3(33%). Isolates from infiltrative form of TB were either N2 (40%) or non clusterisated isolates(40%). It was interesting to mention that 90% drug-resistant isolates belong to some groups of clusters but not the one from drug sensitive isolates belong to any group of clusters.
CONCLUSION We conclude that IS6110-PCR method of TB typing give wide abilities to investigate TB epidemiology. And our preliminary data shows high level of clusterisation of isolates from Novosibirsk. It indicates that level of transmission on TB infection in Novosibirsk is very high, especially, level of transmission of drug-resistant strains.
