Retinoid Receptors levels in normal bronchial mucosa and in lung cancer cell lines

Abstract Lung cancer is a public health problem for which early screening remains in infancy.However,we reported that Retinoid Receptors (RR) expression is down regulated in lung cancer precursor lesions,affecting mainly RARβ.It is our goal, to try to restore a normal RARβ expression in the bronchial mucosa of patients at risk to develop lung cancer.Second generation retinoids,aerosolysed on site,could be used but we need to able to quantify RR.We therefore measured the normal bronchial mucosa level of each RR mRNA by quantitative RT-PCR by LightCycler .RR primers and actin were chosen for their capacity to work at the same pairing temperature;actin being standard.Fourteen lung cancer cell lines were also screened in the same way.In some cell lines, isolated RR mRNA abscence or decreased levels were observed, the levels of RR protein, as measured by Western blot, were not corresponding.RR promoter methylation was also accompanied by unconstant disappearance of the RNA messages.We conclude, that measuring RR mRNA levels is not enough to appreciate RR expression,RR protein levels must be appreciated.This study indicates that RR silencing is also due to several different mechanisms like RR promoter methylation, chromatin compaction or RR protein overdegradation.
