MMP-2 as a marker of human lung carcinoma cells

Abstract

Structural changes in the extracellular matrix (ECM) are necessary for cell migration during tissue remodeling and tumor invasion. The matrix metalloproteinases (MMPs) and their inhibitors have been shown to be critical modulators of ECM composition and are thus, crucial in neoplastic cell invasion and metastasis. In this study, we investigated the expression of MMP-2 in primary mesenchymal lung cells (fibroblasts and vascular smooth muscle cells), lung tumour cell lines (A549, Calu3, H1155, H1299) and in a human bronchial epithelial (HBE) cell line. Interestingly, we found that MMP-2 expression and activity were dominant in the mesenchymal and tumour cell lines while a dominant MMP-9 expression was restricted to the HBE cell line. We also found by zymographic analysis that MMP-2 activity in human lung tumour biopsy (n= 7) was higher compared to healthy parts of the lung from the same patient. Our results suggest that the expression of MMP-2 which is very low in epithelial cells is switched on during the transformation of a normal epithelial cell into lung tumour cells.
