Alterations in MyoD, myogenin and myosin heavy chain expression during mechanical ventilation in rats

Abstract

Since MyoD/myogenin may play a role in fiber phenotype adaptation and MyoD deletion is associated with decreased diaphragm force,the expression of MyoD,myogenin and myosin heavy chain isoforms (MHC) were examined in the diaphragm of controls (C),anesthetized spontaneously breathing rats-24h (SB),anesthetized and MV rats-24h. MV decreased mRNA of MyoD (-56%,p<0.001 vs C,p<0.05 vs SB),MHC-2a (-20%,p<0.001 vs C) and 2b (-21%,p<0.05 vs C) while it increased myogenin mRNA (+67%,p<0.01 vs C). In the SB rats,only MyoD (-33%,p<0.01 vs C) and MHC-2a mRNA (-10%,p<0.05 vs C) decreased.MyoD and myogenin protein expression followed the changes at the mRNA. MHC did not change.Whether the effects of MV on the diaphragm were due to deconditioning or to rhythmic passive shortening was examined on the gastrocnemius.The changes in the 2 muscles followed a similar pattern except for MyoD protein which increased in the gastrocnemius and decreased in the diaphragm while its mRNA diminished.Passive shortening produced no additional effects compared to deconditioning.The alterations in MyoD expression may be involved in impaired diaphragm function seen after 24hMV.The role of deconditioning and passive shortening in these alterations could not be fully unravelled.
