Simultaneous mRNA expression analysis of transforming growth factor β1 and three types of TGF-β receptors in broncho-alveolar lavage cells from patients with interstitial lung diseases – the pilot study

Abstract The aim of the study was to analyze, simultaneously the expression of mRNA of TGF-β1 and 3 types of TGF-β receptors in BAL cells obtained from patients with different interstitial lung diseases (ILD). The group of 18 patients consisted of: 5 pts with idiopathic pulmonary fibrosis (IPF), 3 pts - extrinsic allergic alveolitis (EAA), 5 pts - sarcoidosis I° stage, 5 pts - sarcoidosis II° stage. None of them have been treated before. The control group – three healthy volunteers. In all subjects BAL was performed. From the cell suspension the real-time Q-RT-PCR for mRNA expression of TGF-β1 and his receptors (R-I, R-II, R-III) was performed using ABI PRISMÔ 7700 analyzer. The highest expression of TGF-β1 mRNA was noted in sarcoidosis II° stage, next in sarcoidosis I° stage and in the IPF (log10=3.41>1.29>0.79 copies/μg of total RNA). No expression of TGF-β1 mRNA was observed in the EAA group but in the control group - 0.56 copies/μg. Analyzing the mRNA expression of TGF-β1 receptors’ in BAL cells, three patterns were distinguished: the control group and sarcoidosis I° stage: R-II, R-I, R-III; IPF patients: R-I, R-II, R-III; sarcoidosis II° stage and EAA: R-I, R-III, R-II. BAL cells in sarcoidosis II° stage are important source of TGF-β1. Altered patterns of receptors’ expression in TGF-β system suggest differentiated mechanisms of activation of the immune system cells participating in alveolitis phase of evaluated ILD.
