Abstract

Apoptosis can be stimulated or inhibited by different signals or cellcycle arrest proteins. Some of these regulators are proapoptotic and some are antiapoptotic; each plays a specific role during Mycobacterial infection. This study analyses the expression and corelation of apoptotic and antiapoptotic proteins (P53, FAS, BAX and BCL-2) in human TBgranulomatous reactions.
Immunohistochemicalstaining of 28, 26, 12, 32 formalinfixed paraffinembedded blocks of different biopsy specimens from 40documented TBpatients were studied for expression of P53, FAS, BAX and BCL-2 proteins, respectively. In epitheloidmacrophages of TBgranulomas, high positivity for P53 (100%), BAX (83.3%) and FAS (57.7%) ,was associated with downregulation of BCL-2 (12.5%). Although, the reaction of surrounding small lymphocytes were viseversa. The multinucliatedgiant cells also revealed positive reaction for p53 (92.8%) and BAX (83.3%) expression.
Our studied revealed corelation between highexpression of apoptotic factors; P53, FAS and BAX with downregulation of BCL-2 in epitheloidcells of granulomatous tissue reactions in TBpatients. Our study also reemphasize the value of immunohistochemicalstudies to clarify more detailed information about the role of apoptosis in TBpathogenesis. These studies may lead to further effective diagnostic modalities and better therapeutic strategies in TBpatients.
