Abstract
Objectives:
Early diagnosis of MTB infection is important for both the successful treatment and prevention of disease progression. The microbiologic confirmation of MTB is often difficult and during last years
in vitro
immunodiagnostic tests acquire great importance. Certain highly immunogenic secreted antigens of
M. tuberculosis
such as ESAT-6 and CFP-10 offer hope for more specific diagnosis of MTB infection.
Methods:
10 TB+ and 10 infected with MTB/HIV-1 patients were studied before therapy and 6 months later. ELISPOT (reagents were provided by Oxford Immunotec) and ELISA (QuantiFERON-TB Gold) assays that detect T cell-mediated IFN-gamma responses to ESAT-6 and CFP-10 proteins were used.
Results
: At diagnosis both TB+ and TB+/HIV+ patients had positive results in ELISPOT and in ELISA. After six months responses to ESAT-6 and CFP-10 were still detectable only in one TB+/HIV+ patient who had microbiological isolation and absence of clinical improvement.
Conclusions
: The results obtained stimulated us to conclude that the
in vitro
immunodiagnostic assays could be successfully used for monitoring the efficacy of antituberculosis therapy in TB+ and TB+/HIV+ patients.
