The dynamic of pH, hydrogen peroxide and nitrite in exhaled breath condensate from patients with COPD and asthma exacerbation

Abstract

The analysis of exhaled breath condensate (EBC) biomarkers is non-invasive method for evaluation of the oxidative stress and inflammation in the respiratory tract of patients with pulmonary diseases.
The aim: To evaluate the dynamic of pH-level, the concentration of hydrogen peroxide (H2O2) and nitrite (NO2-) in EBC from patients with COPD and asthma exacerbation before and after 14 days treatment.
Methods: We enrolled 28 patients (14 – COPD and 14 – asthma) and 14 healthy nonsmokers. EBC was collected using ECoScreen before and after 14-days treatment. pH level was measured using Beckman microelectrode. H2O2 concentration in EBC was detected using Prussian Blue based sensor. NO2- concentration in EBC was measured by modified Griess reaction. Spirometry was evaluated also.
Results: The pH was decreased and H2O2 was increased in EBC from patients with COPD and asthma exacerbation in compared with healthy subjects (p<0.05). NO2- in EBC was decreased in all groups of patients, but significant difference was only in the group of asthma compared with healthy (p=0.005). After treatment there were significant increase of pH and decrease of H2O2 in EBC from patients with COPD and asthma, but NO2- wasn‘t significant changed. Negative correlation was found between H2O2 and NO2- in EBC from asthma patients (r=-0.64). No correlations were found between dynamic of biomarkers in EBC and dynamic of lung function parameters.
Conclusion: Determination of EBC biomarkers helpS to assess inflammatory activity in the respiratory tract of patients with COPD and asthma exacerbation and effect of treatment.
