The proton pump inhibitor, lansoprazole, inhibits rhinovirus infection in the cultured human tracheal epithelial cells

Abstract Introduction The entry of major type rhinovirus, type 14 rhinovirs (RV14) into the cytoplasm of infected cells is mediated by the destabilization from receptor binding, by endosomal acidification, or both. Proton pump inhibitors (PPIs), the inhibitors on H-K ATPase in gastric parietal cells, are related to these mechanism. Therefore, it is expected to inhibit infection. Methods Human tracheal epithelial cells were cultured and was infected with the RV14 (5 log TCID50 units/ml) . Cells were treated with lansoprazole (10 μM) before and after the infection and used to examine the inhibitory effects. Results Lansoprazole significantly decreased the viral titers of the supernatants at day 1 (4.1±0.1 log TCID50 units/ml vs. 3.0±0.3 log TCID50 units/ml, n=5) and day 3 (4.7±0.2 log TCID50 units/ml vs. 3.3±0.3 log TCID50 units/ml, n=5). Lansoprazole also inhibited the ICAM-1 mRNA expression in the human cultured tracheal epithelial cells (0.69±0.11 ICAM-1/GAPDH ratio compared with control, n=3), and reduced the contents of soluble form of ICAM-1 in the supernatants (205±15 ng/ml vs. 148±8 ng/ml, n=3) before RV14 infection. Lansoprazole decreased the number of acidic endosomes and the fluorescence intensity from the acidic endosomes in the epithelial cells. Conclusions These findings suggest that lansoprazole may inhibit RV14 infection by reducing ICAM-1 and by blocking the rhinovirus RNA entry into the endosomes.
