Immune regulatory effect of Mycobacterium vaccae vaccine on asthmatic guinea pig

Abstract

Objective:To study the immune regulatory effect of Mycobacterium vaccae vaccine(M.vaccae) on asthmatic guinea pig.
Method:30 guinea pigs were randomly divided into NS,asthma and M.vaccae groups.A guinea pig model of asthma was formed with ovalbumin.Every guinea pig of M.vaccae group was injected intramuscularly with 22.5µg M.vaccae 10 days before ovalbumin immunization.Pulmonary function (Re,resistance of expiration) and airway hyperresponsiveness(PC20,the provocative concentration of inhaled methacholine aerosol required to increase Re value by 20% from the baseline),total IgE and ovalbumin-specific IgE in serum,eosinophils in bronchoalveolar lavage fluid (BALF), mRNA expression of IL-4,IL-5 and IFN-γ in lung tissue of every guinea pig were determined after the last challenge.
Results:Compared with asthma group,M.vaccae group had a significant decrease of Re (p<0.01),a significant increase of PC20(p<0.05),a significant decrease of total IgE (p<0.05) and ovalbumin-specfic IgE(p<0.05)in serum,eosinophils (p<0.01) in BALF, mRNA expression(OD value) of IL-4(p<0.05)and IL-5(p<0.05), a significant increase of mRNA expression of IFN-γ(p<0.05) in lung tissue.
Conclusion:Our results showed M.vaccae could prevent the development of asthmatic guinea pig by regulating Th1/Th2 immune response in lung tissue and decreasing the level of IgE in serum.
